Exosomes, nano-sized vesicles ranging from 30 to 200 nm, are continuously released by nearly all cells. Their capability to traverse cellular boundaries and deliver lipids, proteins, and nucleic acids makes them a promising cell-free therapeutic option for various diseases.

In this study, we explored for the first time whether exosomes derived from human adipose tissue mesenchymal stem cells can alleviate atopic dermatitis (AD) in an in vivo mouse model.

When administered either intravenously (IV) or subcutaneously (SC) to NC/Nga mice treated with house dust mite antigens, ERC-exosomes significantly reduced pathological symptoms, including clinical scores, serum IgE levels, blood eosinophil counts, and the infiltration of mast cells, CD86+, and CD206+ cells in skin lesions. Additionally, ERC-exosomes markedly decreased mRNA expression of inflammatory cytokines such as interleukin (IL)-4, IL-23, IL-31, and tumor necrosis factor-alpha (TNF-α) in AD skin lesions of Nc/Nga mice. These findings indicate that ERC-exosomes could be a novel, promising cell-free therapeutic modality for treating AD.
